Ouachita · Multiplication · Working Protocol

Know what matters before you know to ask.

MyTCLab now explains why a field matters, separates your working protocol from published evidence, and chooses measurement tools from what you actually own.

What the evidence says: the current BAP/IBA working targets sit inside concentrations reported in blackberry multiplication studies, but the reviewed sources did not provide an exact Ouachita-specific media recipe. MyTCLab therefore keeps your formula as a documented working protocol instead of calling it “canonical Ouachita.”

What to check on your next multiplication observation

Measurement readiness

Plants in Jars BAP: 1 mg/mL

Plants in Jars IBA: 1 mg/mL

BKMLAB P200: 20–200 µL

Joanlab P1000: 100–1000 µL

Prepare

Scale from final scientific targets, not from naked stock volumes. The app derives stock draws from the saved supplier concentration and then selects a suitable pipette.

Working amount only. Verify against the exact MS product label.

Ouachita · Multiplication

Full-strength MS working profile · Sugar 30 g/L · Agar 8 g/L · BAP 1.0 mg/L · IBA 0.1 mg/L · pH 5.7–5.8

Evidence status: User working protocol within published blackberry ranges; exact Ouachita-specific validation was not located in the reviewed sources.

Dilution Helper

Use this only when a direct required draw is below the practical range of every saved tool or when the protocol explicitly requires a working dilution.

Measurement Assistant

Enter a liquid draw and MyTCLab checks your saved pipettes. It prefers a direct measurement when one fits.

Pipette Coach

1 · FIRST STOPPress to the first stop before entering the liquid.
2 · ASPIRATESlowly release the plunger while the tip is immersed.
3 · DISPENSEPress to first stop, then continue to second stop for blowout.
4 · RESETWithdraw appropriately, then return the plunger to ready position.
Do not aspirate from the second stop during ordinary forward pipetting. Always follow the manufacturer instructions for your exact pipette and liquid type.

Advanced / legacy calculator

The original Sowa48 calculator is preserved for comparison while V10 extracts its strongest logic into this evidence-centered workflow.

Batches

Track each run from its parent lineage through stage and location changes. Transfers and stage changes append dated history instead of erasing earlier state.

Create batch

Cultures

Manage culture-level identities, containers, derived lineage, transfer history, and check status. Culture records sync through the account’s versioned records.

Locations

One synchronized hierarchy powers the fast list, 2D rack map, and interactive 3D visualization. Move records from any view; every move is logged.

Protocols

A protocol is more than a list of ingredient volumes. Each stage now carries evidence class, scientific targets, working values, and known gaps.

Research & Evidence

The software tells you what is established, what is generalized, what is your own working practice, and what still needs validation.

Cultivar
Ouachita — University of Arkansas cultivar; commercial tissue-culture production is documented.
Exact Ouachita recipe
Not located No peer-reviewed cultivar-specific media formula was located in the V10 research pass.
Blackberry framework
Strong genus-level evidence A 2025 comprehensive review summarizes more than five decades of blackberry micropropagation research.
Multiplication range
BAP was reported across a broad 0.4–10 mg/L range and IBA around 0.1–1.2 mg/L in the reviewed multiplication literature; the review's generalized operational framework highlights BAP 2–5 mg/L with IBA 0.1–0.5 mg/L.
Your current working target
BAP 1.0 mg/L + IBA 0.1 mg/L. This is within the broader published blackberry ranges, but it is retained as a user working protocol, not relabeled as Ouachita-validated.
Rooting framework
Full, half, and other MS strengths have been reported; IBA 0.1–2 mg/L is common in the review. Half-strength MS + IBA 1 mg/L is research-compatible.

Equipment & Pipette Coach

The software chooses a tool from the required draw. It does not invent a dilution merely because a number looks small.

P200

BKMLAB 20–200 µL

Best-fit range for direct draws from 20 to 200 µL. Example: 56 µL fits this range directly.

P1000

Joanlab 100–1000 µL

Best-fit range for larger direct draws. Example: 280 or 560 µL fits in one draw.

RULE

No automatic 1:10

Dilution appears only when a required direct draw is below all saved tools' practical ranges and compatible diluent information is confirmed.

Forward pipetting — ordinary aqueous solutions

1 · FIRST STOPPress only to the first stop before the tip enters the liquid.
2 · ASPIRATEImmerse the tip and slowly release the plunger to draw the selected volume.
3 · DISPENSEPress to the first stop, then continue to the second stop to blow out the remaining liquid.
4 · RESETWithdraw according to technique, then let the plunger return to ready position.
Why first vs second stop matters: the first stop corresponds to the calibrated selected volume in forward pipetting. The second stop is the blowout stroke used during dispensing. Do not aspirate from the second stop when using ordinary forward pipetting.

Always follow the actual pipette manufacturer's instructions and never dial beyond the stated range. Accuracy and precision depend on the instrument, calibration, tips, technique, temperature, and liquid properties.

Guided Lab Notebook

Instead of “take notes,” MyTCLab asks for information that becomes valuable later and tells you why.

Observation discipline

Observation

What did you actually see or measure?

Interpretation

What do you think it may mean?

Action

What did you do because of it?

Outcome

What happened afterward?

Saved observations

Acclimatization / Hardening Off

This is modeled as a protocol checklist with readiness gates, not as a fake “formula.”

Your four-week schedule can be saved as a local working plan, but the software will not imply that every Ouachita plantlet must advance simply because a calendar week changed.

Readiness to leave culture

Root development, shoot quality, contamination status, morphology, and protocol criteria.

Humidity transition

Record dome/cover state, ventilation, visible wilt, new growth, and survival over time.

Substrate & environment

Record substrate, container, light transition, watering, temperature context, and when protection is reduced.

Tissue Culture Academy

When a field or warning is unfamiliar, the software should be able to teach the concept instead of making a beginner leave the workflow.
