LEVEL 05

Culture Lifecycle

Understand the changing objective across initiation, multiplication, rooting, transfer, and acclimatization.

01

Initiation

Initiation or establishment is the stage in which selected source material is introduced into culture and an acceptable starting culture is established. The work focuses on identity, contamination management, viability, and early response rather than rapid multiplication.

02

Multiplication

During multiplication, the objective is to increase useful propagules, commonly shoots, while maintaining acceptable morphology and identity. Subculture timing, plant growth regulators, vessel environment, and genotype can all affect multiplication quality.

03

Rooting

Rooting prepares suitable shoots or plantlets for root development where the propagation system requires it. Some protocols root in vitro; others use ex-vitro rooting or combine rooting with acclimatization.

04

Acclimatization

Acclimatization transitions plantlets from the protected, humid, sugar-supported in-vitro environment toward ex-vitro conditions. In-vitro plants can have altered anatomy and physiology, making the transition a genuine biological stage rather than simple repotting.

05

Transfers and subculture

Subculture moves material to fresh culture conditions or divides it into derived cultures. Each transfer can change lineage, vessel count, medium context, contamination exposure, and developmental state.

06

Stage transitions

A stage transition should be based on biological readiness and the protocol's criteria, not only on elapsed time. Useful criteria may include shoot development, rooting status, morphology, contamination status, or other plant-specific observations.

07

Lifecycle simulation

A lifecycle simulation asks the learner to follow one culture record from source plant through stages, transfers, and final acclimation while keeping identity, recipe version, location, and observations attached.
