A common teaching model for micropropagation is establishment/initiation, multiplication, rooting, and acclimatization, often preceded by donor-plant preparation. Real protocols may combine, omit, or modify stages, but the model helps beginners understand that the objective changes over time.

Understand it

Describe the usual objectives of culture stages

Initiation aims to establish appropriate plant material in culture.

Initiation or establishment focuses on obtaining a suitable starting culture rather than maximizing multiplication.

Distinguish multiplication from rooting and acclimatization

Multiplication aims to increase usable propagules or shoots.

Multiplication aims to increase useful propagules, while later work may focus on rooting and adjustment outside culture.

Use the stage model without forcing every protocol into it

Rooting and acclimatization prepare plantlets for later growth outside the in-vitro environment where appropriate.

Real protocols may combine or modify stages, so record what actually happened rather than forcing a standard diagram.

Worked example

A record follows source preparation, establishment, multiplication and then a protocol that roots shoots outside the vessel. The grower records the actual path instead of inventing an in-vitro rooting event simply to fill every box in a diagram.

Explain your decision before opening the worked reasoning below.

Show the worked reasoning

The stage model organizes objectives; it is not evidence that every plant follows an identical sequence or schedule.

Acclimatization addresses the transition toward ex-vitro growth while preserving the culture's earlier identity and history.

Important boundary

Use the stage model as a map, not as proof that every species follows an identical route.

Try it

How should the example's ex-vitro rooting be recorded?

Use the worked example above to explain your answer in your own words before taking the quiz.

Lesson quiz

Assessment version: 2026-10-15-v2. Previous reading-completion markers are retained. This revised quiz has its own result.

Apply it

Use the worked example to write a short decision record: what did you observe, which rule did you apply, and what would you check before acting?

Completion marks reading progress, not a quiz pass or professional certification.
